How does the StripDetective test work?
StripDetective is a presumptive screening test designed to detect elevated acid phosphatase activity—an enzyme found at particularly high concentrations in seminal fluid. For this reason, acid phosphatase testing has long been used in forensic science as a preliminary method for identifying biological traces that may contain semen.
The reaction on the test area takes place in two stages. First, acid phosphatase catalyses the removal of a phosphate group from α-naphthyl phosphate, which acts as the reaction substrate. The resulting α-naphthol then reacts with a diazonium salt to form a coloured compound, producing the characteristic violet or purple appearance of the test area.
The development of a violet colour within the specified reading time indicates that acid phosphatase activity has been detected above the test’s detection threshold. Higher enzyme activity may result in faster colour development or a more pronounced reaction.
However, the StripDetective formula contains more than these two reactive compounds. Test also includes pH-buffering agents, stabilisers, protectants, reaction enhancers and inhibitors designed to suppress non-specific phosphatase activity. Together, these components provide favourable conditions for the enzymatic reaction, improve reagent stability, enhance the visibility of the colour change and reduce the risk of non-specific reactions. As a result, the test is designed to detect low levels of enzyme activity while maintaining a high degree of reaction selectivity.

Instruction of use: how to detect with Strip Detective?
Fill the attached bottle with water. Spray the suspicious stain 2 times.
Spray the test pad 1 time and press it to the stain. The 1cm test pad should be facing the stain.
Leave the test pad on the stain and wait for 1 minute.
Check the result. If the color of the test pad will turn purple - the stain contains acid phosphatase. Please do the profesional laboratory test.

If you are not sure about the final result, you can do the negative control.
Please take unused strip and spray it with water only. Wait for 1 minute and compare the control result with your result. If the negative result color will be the same as yours – interpret it as negative too.

What it means if the purple color is less or more intense?
More intense colour indicates a higher concentration of acid phosphatase in the trace (and high concentration of semen). Sperm contains high concentrations of acid phosphatase. The concentration of semen in the trace may change due to amount od water added, the destruction of the trace during washing, time influence effect and others.
If the test is positive you should follow it with certified laboratory procedure.
What is the expiry date of Strip Detective?
The test can be used for one year from the date of manufacture. The expiry date is printed on the box.

How to interpret the results
Positive result for semen detection
The development of a purple colour on the test area after 1 minute indicates that acid phosphatase activity has been detected. The intensity of the colour may depend on the level of enzyme activity in the tested sample.
Compare the colour of the test area with the reference colour scale printed on the packaging. If, after 1 minute, the colour corresponds to the positive result shown on the scale or is more intense, the result should be interpreted as presumptive positive.
A presumptive positive result indicates elevated acid phosphatase activity and should be verified by an accredited forensic laboratory to confirm the presence of semen.
Negative result for semen detection
The absence of purple colour, or a colour reaction below the threshold shown on the reference scale, should be interpreted as a negative result. This means that the test did not detect acid phosphatase activity above its detection threshold. A negative result does not completely exclude the presence of semen, particularly when the trace is small, diluted, degraded or incorrectly collected.
Important: A positive chemical test result does not provide definitive confirmation that semen is present. Acid phosphatase may also occur at lower levels in other biological materials. A presumptive positive result should therefore be confirmed by laboratory analysis performed by a qualified forensic expert.
Read the result exactly 1 minute after performing the test. Colour changes that develop after the specified reading time should not be used when interpreting the result.
